III.7. Reusability of immobilized α-amylase

The main advantage of enzyme immobilization is that it leads to a reusable enzyme preparation. Reusability of enzyme is a key factor for its cost-effective industrial use. Immobilized α-amylase in calcium alginate beads can be reused either by filtration or centrifugation. The reusability of immobilized α-amylase was studied up to 10 cycles. As shown in Fig.15, the activity of immobilized α-amylase decreased to approximately 35% of its original activity over ten cycles.

CONCLUSION

1.The influence of various factors on the growth of Bacillus subtilis and the process of α-amylase production was studied. Extracellular α-amylase production and growth of Bacillus subtilis was maximal at pH 7 and the optimal temperature for production of α-amylase and growth of B. subtilis occurred at 45°C.

2.The best carbon source for α-amylase production from B. subtilis was found to be starch and the optimal production of α-amylase and growth of Bacillus subtilis were obtained when peptone was used as nitrogen sources.

3.The maximum starch concentration for the optimal α-amylase production from Bacillus subtilis was 1.0%. α-amylase production and growth of Bacillus subtilis increased after 24 to 48 h. After this period, the production of the enzyme decreased.
