2.6 Other analytical methods and statistical analysis

 

Chemical  oxygen  demand  (COD)  and  phosphate  levels  were  determined  based 

on  the  standard  methods  advocated  by  the  American  Public  Health  Association

(APHA) (APHA, 1998). The PHA and glycogen assays were carried out according to 

a  previously  described  protocol  (Oehmen  et  al.,  2005).  The  activities  of 

exopolyphosphatase  (PPX)  and  poly-P  kinase  (PPK)  were  measured  using  the 

methods described by Chen et al. (2012). Reactive oxygen species (ROS) production 

and  extracellular/intracellular  Ti  distribution  were  determined  using  the  methods 

described  in  our  previous  report  (Li  et  al.,  2019a).  The  ultrastructures  of  the  sludge 

flocs were examined and analyzed by transmission electron microscopy (TEM) using 

an  energy-dispersive  X-ray  spectrometer  (EDS),  and  the  elements  present  in  the 

bacterial cells found in the corresponding sludge were identified. We determined the 

EPS  contents  and  assessed  the  release  of  lactate  dehydrogenase  (LDH)  using  the 

methods described in our previous report (Li et al., 2019b). 

The experimental values were compared to their corresponding control values for 

the  purposes  of  statistical  analysis.  We  assessed  the  significance  of  the  results  by 

performing  a  statistical  analysis  test  (t-test),  and  p-values  <  0.05  were  considered 

statistically significant.
