2.4 DNA extraction and Illumina MiSeq sequencing

 

After  the  exposure  experiments,  sludge  samples  (0.5  g  each)  were 

homogeneously harvested from each SBR system, and immediately stored at −80 °C 

until  required  for  DNA  extraction.  The  DNA  extraction  and  Illumina  MiSeq 

sequencing  were  strictly  in  accordance  with  our  previous  research  (Li  et  al.,  2019a) 

and the analysis methods used by Hu et al. (2017). Each DNA sample was extracted

using  an  E.Z.N.A.  Soil  DNA  kit  (D5625-01;  OMEGA,  USA)  according  to  the 

manufacturer’s  instructions.  The  concentrations  of  the  extracted  DNA  were 

determined  by  0.8%  (w/v)  agarose  gel  electrophoresis.  Polymerase  chain  reaction 

(PCR)  amplification  was  performed  using  the  primers  515F  (5′-GTGCCAGC 

MGCCGCGG-3′)  and  907R  (5′-CCGTCAATTCMTTTRAGTTT-3′).  We  sent  the 

DNA  samples  to  Guangdong  Magigene  Biotechnology  Co.,  Ltd.,  China  for  high 

throughput sequencing analysis.
