2.3 Extraction and species analysis of P in sludge cells and EPS

 

During  the  last  cycle  after  exposure,  we  obtained  100-mL  samples  of  the 

uniformly  mixed  activated  sludge  suspension  at  0,  2,  and  6  h.  The  extraction  of  P 

species  from  the  sludge  cells  and  EPS  was  based  on  a  slightly  modified  previous 

protocol (Xu et al., 2017). Briefly, a cation exchange resin (CER) was used to extract 

the P species from the EPS in the 100-mL sample of activated sludge. We then used 

cold  perchloric  acid  (PCA)  fractionation  combined  with  NaOH  extraction  to 

fractionate  the  P  species  in  the  sludge  residue  cells  from  CER  extraction.  The 

detection  of  total  phosphorus  (TP),  orthoP,  and  complex  phosphorus  (defined  as 

TP-orthoP)  in  the  extraction  solution  mentioned  above  was  based  on  the  reported 

definitions (Xu et al., 2017).
