A nnotation.
Allochrusa gypsophiloides(local
name Beh, Etmak) is the best known endemic saponin-
bearing plant of Central Asia. Ultra-high performance liquid chromatography-electrospray ionization-mass
spectrometry (UPLC-ESI-MS) was applied to the analysis of the compounds of the roots of
A.
gypsophiloides.
Through UPLC-ESI-MS studies on the plant methanol extract triterpene saponin -3-O-p-D-

Introduction
Allochrusa
is a genus of family Caryophyllaceae comprising about 8 species distributed in Turkey,
Central Asia, Afghanistan, Caucasus, and Iran. One species of
Allochrusagypsophiloides
occurs in Central
Asia.
A. gypsophiloides
has a relatively limited area, it grows only in the western Tien Shanand Pamir-Alay
at foothill desert loess steppes, dry slopes of rivers, on gravelly slopes at analtitude of 400-1300 meters
above sea level. This species has been known as a valuable source of saponin on the territory of the
republics of Central Asian countries such as Uzbekistan and Kazakhstan, has been exported for a long
time. Annual planned volumes of collecting dry roots reached 700 - 800 tones [1]. As a result of the
intensive and unsystematic harvesting of
A. gypsophiloides
brings it to the list of rare species with a strongly
decreasing population and recorded in the Red Book of Uzbekistan [2].
Triterpene glycosides are natural low molecular weight compounds that are widespread in plants
and some sea animals. Among them
Allochrusa
are a rich source of triterpene glycosides with diverse
chemical structures and physiological activities.Although, there are some reports about glycosides
(triterpene saponins) in this plant [3, 4],UPLC-ESI-MS investigations have not yet been conducted on
A.
gypsophiloides.
Therefore, the objectives of this study were to study saponins of the methanol extract of
A.
gypsophiloides
using UPLC-ESI-MS. Herein, we report the known saponin in the methanol extract from
A.
gypsophiloides
roots.
Results and discussion
Liquid chromatography coupled to mass spectrometry (LC-MS) is an excellent technique to analyse
multi-components in complex herbal matrices. Due to its inherent characteristics of accurate mass
measurements and high resolution, time-of-flight (TOF) MS is well-suited to this field, especially for
qualitative applications[5]. Roots from
A. gypsophiloideshave
been shown to accumulate triterpenoid
saponins [3, 4]. They belong to the group of glucuronide quillaic- and gypsogenin type triterpenoid
carboxylic acid 3,28-O-bidesmosidessaponins. Compounds from this group have a glucuronic acid moiety
at C-3 hydroxyl of the aglycone and are among the highest glycosylated bidesmosides [6]. Negative ion
ESI-MS of saponins offers high sensitivity and more diagnostic fragment ions compared to positive ion MS.
